Go to the Services tab and scroll to look for the Windows Defender service (Check the box) and then go to the Startup tab and then look for Windows Defender (Check the box) and click OK.
This will prompt you to restart your machine. Once the machine boots back, your computer should now turn on Windows Defender during startup.

Just click on Start > Run and type MSCONFIG

Once you have turned on Windows Defender, it should remain on until you turn it off again, regardless.
